What is wrong / is it possible to fix two pixel-columns on the 48sx screen? They are blank. The rest of the screen works ok.

It is usually (~98% of the time) a damaged LCD. The replacement can come from any older 38, 39 or 48 machine. That is to say the 48Gii, 49G+, 39G+ and others made in China are not compatible.
